I assume you are referring to the automatic pausing of the AuraDB Free instances after 72 hours of no activity, if not, please correct me.

Submitting a Query before you stop work on a Friday, maybe programatically via cron, would mean that the instance is still up and running on your return, where you can then run another query against the database.

Note AuraDB Professional instances do not auto pause after periods of inactivity.
